The goal of this study was to characterize the spatial, seasonal and annual hepatic activities of mixed-function oxidase (MFO) in the speckled sanddab Citharichthys stigmaeus, the most common fish in the Moss Landing area. In addition, techniques to monitor MFO activities in caged speckled sanddabs were developed and tested. Once the relationship between MFO activities in caged and wild fish populations is determined, caged fish could be used to monitor potential hydrocarbon impacts at Moss Landing, or other marine sites. During each of the spatial, seasonal and annual sediment samplings conducted in 1985-1987 as part of a separate hydrocarbon variability study at Moss Landing, 12 wild speckled sanddabs were collected from Moss Landing Harbor, Elkhorn Slough and nearshore Monterey Bay sites. In addition, four locations were chosen for a 14-day field caged fish experiment. The caged fish experiments successfully demonstrated the feasibility of using caged sanddabs as indicators of hydrocarbon exposure. The major source of variability in hepatic aryl hydrocarbon hydroxylase activity in wild speckled sanddabs from the Moss Landing area is due to seasonal rather than site differences. Significant relationships between caged fish MFO response and sediment hydrocarbon concentrations were found. It is possible that caged fish could be used in place of costly sediment sampling and analysis, and provide a more direct method to assess biological impacts. Many of the caging techniques demonstrated in this study can easily be transferred to other benthic flatfish, and other marine and freshwater environments.  相似文献

Metal phytoextraction with hyperaccumulating plants could be a useful method to decontaminate soils, but it is not fully validated yet. In order to quantify the efficiency of Cd and Zn extraction from a calcareous soil with and without Fe amendment and an acidic soil, we performed a pot experiment with three successive croppings of Thlaspi caerulescens followed by 3 months without plant and 7 weeks with lettuce. We used a combined approach to assess total extraction efficiency (2 M HNO3-extractable metals), changes in metal bio/availability (0.1 M NaNO3-extractable metals and lettuce uptake) and toxicity (lettuce biomass and the BIOMET biosensor). The soil solution was monitored over the whole experiment. In the calcareous soil large Cu concentrations were probably responsible for chlorosis symptoms observed on T. caerulescens. When this soil was treated with Fe, the amount of extracted metal by T. caerulescens increased and metal availability and soil toxicity decreased when compared to the untreated soil. In the acidic soil, T. caerulescens was most efficient: Cd and Zn concentrations in plants were in the range of hyperaccumulation and HNO3-extractable Cd and Zn, metal bio/availability, soil toxicity, and Cd and Zn concentrations in the soil solution decreased significantly. However, a reduced Cd concentration measured in the third T. caerulescens cropping indicated a decrease in metal availability below a critical threshold, whereas the increase of dissolved Cd and Zn concentrations after the third cropping may be the early sign of soil re-equilibration. This indicates that phytoextraction efficiency must be assessed by different approaches in order not to overlook any potential hazard and that an efficient phytoextraction scheme will have to take into account the different dynamics of the soil-plant system.  相似文献

Nitrifier growth parameters--the maximum growth rate (microAmax) and yield (YA)--were estimated by measuring the rate of carbon dioxide uptake and additional rates of oxygen uptake and ammonia (or nitrite) use. Batch tests in a combined titrimetric and offgas analyzer with enriched Nitrobacter and Nitrosomonas cultures and an activated sludge sample were performed. The measured microAmax values for the Nitrobacter and Nitrosomonas cultures were 0.67 +/- 0.03 day(-1) and 0.54 +/- 0.09 day(-1), while the YA values were 0.072 +/- 0.01 g volatile suspended solids (VSS) x g nitrogen (N)(-1) and 0.14 +/- 0.02 gVSS x gN(-1), respectively. For the activated sludge sample, microAmax was observed to increase with pH (microAmax = 0.72 x 3.3(pH-7.1)) over the range 6.8 to 7.1. All microAmax and YA values determined by this method were similar to those previously reported. Compared with other microAmax and YA estimation methods, this method allows for unique microAmax and YA estimations for given conditions from a single experiment.  相似文献

Lakes in Killarney Park near Sudbury, Ontario, Canada, have shown dramatic water quality changes including general increases in pH and alkalinity, and decreases in SO4(2-), base cations and metals. While some lakes have recovered to pH > 6.0, many are still highly acidic despite decades of improvement. Very high historical S deposition related to emissions from the Sudbury metal smelters dominated the acidification process in this region. However, since the implementation of substantial S emission controls (90%) at the smelters, the Sudbury emissions are no longer the major source of S deposition in the Sudbury area. Wet deposition of SO4(2-) and SO4(2-) concentrations in lakewaters at Killarney now approach values in the Dorset, Ontario, area, about 200 km from Sudbury. This suggests that the S deposition to the Killarney area is now primarily from long-range transport, not from local sources. Studies of Killarney lakes are revealing the complex nature of the chemical recovery process. As lake acidity decreases, other changes including decreased Ca2+ concentrations, increased transparency, and altered thermal regimes may potentially affect some of these ecosystems. It is clear that continuing assessments of the recovery of Killarney lakes, within a multiple-stressor framework, are needed.  相似文献

Agriculture can be a major nonpoint source (NPS) of nutrient and pesticide contamination in the environment. Available databases do not provide accurate and dynamic data on fertilizer and pesticide application, which limits the ability of complex watershed models to simulate contaminant loads into impaired water bodies. A model for estimating agricultural nutrient and pesticide input for watershed modeling has been developed. Climate, soils, and major agricultural operations are considered within the model, so that it can be adapted to any watershed or subregion within a watershed. The timing of the agricultural operations is a function of the weather data, providing realistic results at daily, monthly, or annual application rates. The model also predicts irrigation demand and biomass production, which can be used to calibrate the model. Model output can be used in any watershed model that considers agricultural land uses. Two case studies were evaluated, using grape vineyards in the Napa River and strawberry production in Newport Bay as examples. The predicted time to maturity corresponded well with actual data. Irrigation and fertilizer needs were very sensitive to weather input. Although the model can generate weather from long-term averages, the simulated results are best when at least observed precipitation and temperature are provided, to capture extreme events. The model has data for 98 crops and 126 pesticides, based on the California Department of Pesticide Regulation database. The databases are easily modifiable by the user to adapt them to local conditions. The output from AgInput is much needed for watershed modeling and for development of total maximum daily loads (TMDLs), based on realistic targets of irrigation, nutrient, and pesticide inputs. An argon ion laser emits several laser lines In the visible region of the optical spectrum. The absorption coefficients of N02 at these laser emissions were measured in a multiple pass absorption cell. A differential technique, in which the ratio of the transmitted intensities of the argon laser emissions is measured, is described to determine the concentration of N02 in a polluted atmosphere over path lengths of several kilometers. Measurement of ratios eliminates interferences from particle scattering and thermal index gradients. Evaluation of the data taken in the 48 meter multipass cell indicates that concentrations of N02 less than one part per million could be determined in a 1 km optical path.  相似文献

A new gas chromatographic method reliable in the microgram range, was developed to measure ETU residues in various crops. Contrary to previously published procedures, this new method does not require prior derivatization of the ETU. The seasonal productivity cycle and factors controlling annual variation in the timing and magnitude of the winter–spring bloom were examined for several locations (range: 42°20.35′–42°26.63′N; 70°44.19′–70°56.52′W) in Boston Harbor and Massachusetts Bay, USA, from 1995 to 1999, and compared with earlier published data (1992–1994). Primary productivity (mg C m−2 day−1) in Massachusetts Bay from 1995 to 1999 was generally characterized by a well-developed winter–spring bloom of several weeks duration, high but variable production during the summer, and a prominent fall bloom. The bulk of production (mg C m−3 day−1) typically occurred in the upper 15 m of the water column. At a nearby Boston Harbor station a gradual pattern of increasing areal production from winter through summer was more typical, with the bulk of production restricted to the upper 5 m. Annual productivity in Massachusetts Bay and Boston Harbor ranged from a low of 160 g C m−2 year−1 to a high of 787 g C m−2 year−1 from 1992 to 1999. Mean annual productivity was higher (mean=525 g C m−2 year−1) and more variable near the harbor entrance than in western Massachusetts Bay. At the harbor station productivity varied more than 3.5-fold (CV=40%) over an 8 year sampling period. Average annual productivity (305–419 g C m−2 year−1) and variability around the means (CV=25–27%) were lower at both the outer nearfield and central nearfield regions of Massachusetts Bay. Annual productivity in 1998 was unusually low at all three sites (<220 g C m−2 year−1) due to the absence of a winter–spring phytoplankton bloom. Potential factors influencing the occurrence of a spring bloom were investigated. Incident irradiance during the winter–spring period was not significantly different (P > 0.05) among years (1995–1999). The mean photic depth during the bloom period was significantly deeper (P < 0.05) in 1998, signifying greater light availability with depth. Nutrients were also in abundance during the winter–spring of 1998 with stratified conditions not observed until May. In general, the magnitude of the winter–spring bloom in Massachusetts Bay from 1995 to 1999 was significantly correlated with winter water temperature (r 2=0.78) and zooplankton abundance (r 2=0.74) over the bloom period (typically February–April). The absence of the 1998 bloom was associated with higher than average water temperature and elevated levels of zooplankton abundance just prior to, and during, the peak winter–spring bloom period.
